Abstract: Single cell RNA sequencing is becoming more commonly used to profile larger numbers of samples, underscoring an important need for an efficient and scalable solution. There is also a growing desire to perform multiomic analysis for more thorough characterization of biological samples. We have developed an assay that enables the extraction of gene expression and protein expression signatures from samples via sequencing, using DNA probes that target the transcriptome and antibody-oligonucleotide conjugates. The workflow is also compatible with paraformaldehyde (PFA) fixation, allowing samples to be collected, shipped to a central location, and processed without sacrificing sample integrity or data quality. This opens new possibilities for sample accessibility, throughput, and batched analysis. Our platform enables profiling of up to a million fixed single cells at once with a scalable workflow that can accommodate various experimental schedules with multiple stopping points or help preserve precious, fragile, and sensitive samples by locking in their cell state(s) with our fixation protocol.
