(1232-A) Automating PacBio® SMRTbell® Prep Kit 3.0 Workflow on the Revvity Sciclone® NGSx G3 Workstation

Abstract: Long-read sequencing is a type of nucleic acid sequencing that uses DNA fragments ranging from 1,000 to 20,000 basepair or more to produce individual reads of thousands of nucleotides in length. Advantages of long-read sequencing include higher accuracy, more uniform coverage, and higher confidence in reconstruction of a genome. An efficient workflow and chemistry used for library prep is essential to producing quality long-read sequencing data. Here we discuss the use of Sciclone G3 NGSx to generate whole genome sequencing (WGS) libraries with the Pacbio SMRTbell Prep Kit 3.0 for sequencing on the PacBio long-read systems. The PacBio® SMRTbell® Prep Kit 3.0 has a streamlined library construction protocol with a gel free workflow by utilizing a bead-based size selection. Combined with Revvity’s Sciclone® G3 NGSx workstation, it offers a high throughput workflow by allowing the user to prepare up to 96 sequencing libraries in one run. Overall yield and sequencing metrics generated with the automated library preparation were within the range of the kit. The automated workflow provides a simple user interface guided workflow, reducing human error, hands-on-time, and overall project costs.
