Abstract: Illumina Complete Long Read Prep with Enrichment Kit uses long-read sequencing technology to access areas of the genome that are traditionally challenging to map. The automation of Illumina Complete Long Read Prep with Enrichment Kit on Revvity’s Sciclone® G3 NGSx Automation workstation generates reliable sequencing-ready NGS libraries in ~2 days. The automation on Sciclone G3 NGSx workstation minimizes the human input by reducing hands-on time, error rate, operational cost and increases walkaway time. Illumina Complete Long Read Prep with Enrichment workflow require a total of 17 steps from Tagmentation to sequencer ready libraries. At the start of each run, the user is prompted to select the step they want to start with and are guided with the images and the text to ensure that the Sciclone G3 NGSx deck is properly setup. Based on the number of columns to run, the workbook provides the master mix calculations and the volume of reagent necessary for each step. A 12-column run with 24 samples using 50ng of Promega input DNA generated high total yield (8.7ng/µl and 13.2nM) and resulted in expected fragment size (~1000bp). The fragment traces confirmed nearly no dimer and the key sequencing metrics were similar to the manual preparation. The Illumina Complete Long Read Prep with Enrichment kit automation on Sciclone G3 NGSx offers a reliable, reduced hand on time and cost saving for generating NGS libraries using high-throughput approach.
