(1265-B) Development of a cross platform IR-MALDESI front end for high throughput ToF Mass Spectrometry

Abstract: Over recent years, AstraZeneca has worked to develop a high throughput mass spectrometry platform which utilizes the speed and contactless nature of acoustics as a sample introduction technology. While the technology has delivered high value to AZ it is not commercially available, therefore has a limited life expectancy within our company. As a result, we continue to develop solutions that can take over from AMI-MS in future. We will share our experiences of collaborating to develop an IR-MALDESI system similar to that published by the group at AbbVie. We have modified the laser optics to improve the laser fluence and illumination of the sample. These modifications focus the laser light as a tight circular spot delivering a high energy beam capable of volatilising the aqueous sample. Our latest prototype has been designed to interface with both the Waters Xevo G2XS and Thermo Exploris 480 time of flight mass detectors. TTP have built a common interface device to enable a quick change over between the two mass spectrometers. With this new device we will be able to extensively test the IR-MALDESI system with a wide range of biological and chemical samples across both detector platforms. Once the initial phase of testing is complete, we will be interested in enabling open access to this technology platform so that scientists outside of AZ can submit and run samples at our state-of-the-art research facility in Cambridge, UK.
