Abstract: Plasmid DNA has many applications and are routinely utilized as bacterial cloning or expression vectors. There is an urgent need for robust time-saving tools for large-scale plasmid DNA purification. While current column-based kits have been widely used for plasmid purification, they also require tedious manual steps, bulky centrifugation or vacuum setups, and leaves behind significant solid and liquid waste. Previous automated purification instruments address these pain points, but still rely on tedious centrifugation, requiring special lab supplies (e.g. in house compressed air, ultra centrifuges), or can only operate in a very narrow bio-input range. Here we showcase a completely redesigned self-contained walkaway instrument enabling maxi-scale plasmid DNA purification without centrifugation in just 75 minutes with less than a 5-minute setup. A solution that integrates a multi-step process into a single cartridge simplifying the purification process into a fully automated instrument offering a one-stop sample to product workflow.
Plasmids are routinely utilized in large scale research applications like protein (e.g., insulin), gene therapies, mRNA therapies, vaccines, genetic engineering, and animal disease research. In 2010, Invitrogen launched the first commercially available plasmid processing station – BenchPro® 2100. Here, we show the next generation of this instrument with increased durability and simplified processes. This walkaway benchtop instrument details robustness, increased capability of processing bacterial culture of optical density (OD600) up to 6, using a “low endo” or premium “endotoxin-free” cartridge. The KingFisher PlasmidPro instrument is a user-centric device with a plug-and-play installation, an intuitive touchscreen and a footprint that fits on a standard lab bench with negligible noise and heat production alongside very low power consumption.
