Abstract:
Introduction: The intestinal microbiome, a diverse ecosystem within the gastrointestinal tract, consists of trillions of co-evolved microorganisms essential for functions like digestion, immune response, and protection against pathogens. Diseases such as inflammatory bowel disease and obesity are associated with imbalances in this community, prompting extensive research. Next-generation sequencing (NGS) has revolutionized microbiome studies, allowing for a thorough analysis of genetic material and insights into previously challenging-to-culture microorganisms, potentially transforming health diagnostics and treatments.
Method: Ortho-Analytic, a Swiss medical laboratory for integrative medicine, aims to make intestinal microbiome analysis more broadly accessible using NGS and automated DNA extraction. Their streamlined workflow, incorporating Calex® NGS tubes, DreamPrep® NAP workstation, and ZymoBIOMICS™ 96 MagBead DNA Kit, significantly reduces collection-to-extraction times, ensuring hands-free operation and providing high-quality DNA for NGS sequencing or qPCR analysis. Tailored for high-throughput labs, it showcases efficiency with reduced production time and increased sample throughput. The integrated Frida Reader™ delivers precise concentration measurements, facilitating direct assessments without material wastage.

Discussion: The reliable substitution of manual with automated extraction is corroborated by excellent correlation in sequencing results, further validated by an external inter-laboratory comparison confirming accuracy at 98.4%.
Conclusions: In summary, this workflow standardizes microbiome analysis, offering simplicity, reducing hands-on time, and minimizing errors, ultimately benefiting researchers and holding the potential for future diagnostic applications.
